Boehringer Ingelheim GmbH’s zongertinib enters the market as the first oral HER2-targeted therapy for patients with non-small-cell lung cancer, following an accelerated approval by the U.S. FDA. Branded Hernexeos, the drug is cleared for use in adults with unresectable disease or whose tumors have HER2 tyrosine kinase domain activating mutations and who have received prior systemic therapy.

Metabolic markers to predict efficacy of immune checkpoint inhibitors in non-small-cell lung cancer

Lung cancer is the second most frequent cancer worldwide, and it accounts for 18% of all cancer-related deaths. Most cases of lung cancer involve non-small-cell lung cancer (NSCLC), in which therapy with immune checkpoint inhibitors (ICIs) can improve prognosis, yet up to 75% of patients fail to respond to it. Researchers at the Chinese Academy of Medical Sciences & Peking Union Medical College have identified potential metabolic markers that may help predict which patients are more likely to respond to ICI therapy.

The use of tyrosine kinase inhibitors (TKIs) in the treatment of epidermal growth factor receptor (EGFR)-mutant non-small-cell lung cancer (NSCLC) has not produced the same durable clinical benefits observed with next-generation targeted therapies in ALK- and ROS1-rearranged NSCLC. For the second time in as many months, the lack of statistically significant overall survival data sent shares of Summit Therapeutics Inc. sliding, despite otherwise positive findings for PD-1/VEGF bispecific candidate ivonescimab in non-small-cell lung cancer. But analysts noted the stock reaction overlooks the consistency of the efficacy data to date, as well as the fact that the phase III Harmoni study marks the first pivotal trial testing ivonescimab in a global population.
