AACR 24: Freezing, of all things, is one way to heat cold tumors

“Hot and cold tumors may need different types of immunotherapy,” Jay Berzofsky told the audience as the American Association for Cancer Research’s (AACR) 2024 annual meeting kicked off this weekend.  In an educational session on cancer vaccines, Berzofsky, who is head of the National Cancer Institute’s Molecular Immunogenetics and Vaccine Research section, explained that when immunotherapy fails in hot tumors, it fails despite the existence of an immune response, due to an immunosuppressive microenvironment. FDA expands Legend, J&J’s Carvykti with ‘best-case’ label in MM

Following the U.S. FDA’s expansion of competing BCMA-targeting CAR T therapy Abecma (idecabtagene vicleucel) to include third-line and later treatment in multiple myeloma (MM) patients, the agency cleared Carvykti (ciltacabtagene autoleucel) from Legend Biotech Corp. and Johnson & Johnson’s Janssen unit for use in MM patients as early as second-line treatment. The label, which RBC Capital Markets analyst Leonid Timashev called a “best-case scenario,” includes no notable updates to the black box warning and should help accelerate and expand Carvykti’s update in the U.S., with 2024 revenues expected to top $950 million. VC firm Medicxi invests $40M in China’s D3 Bio

Medicxi has made its first ever investment in China – and its biggest single commitment to date – putting $40 million into D3 Bio Inc. The investment by the London-based venture capital firm will accelerate development of D3’s lead program D3S-001, a second generation KRAS G12C inhibitor, which is in phase II development in advanced solid tumors. Jury agrees with SEC’s ‘shadow trading’ allegations

A quick jury verdict that a biopharma official was guilty of insider trading validated the U.S. SEC’s broader view of what constitutes such trading and could ignite more SEC “shadow trading” investigations and allegations. Following an eight-day trial before the U.S. District Court for the Northern District of California and a little more than two hours of deliberation, a jury found April 5 that Matthew Panuwat violated national securities laws when he purchased short-term, out-of-the-money stock options in Incyte Corp. in 2016. Ionis eyes olezarsen launch before year-end on final phase III data

The higher of two doses of Ionis Pharmaceuticals Inc.’s drug olezarsen for treating adults with familial chylomicronemia syndrome reduced fasting triglyceride levels by 44% from baseline at the six-month mark in a phase III study, but one case of pancreatitis in the trial paints a less attractive picture than top-line data the firm reported in September. AACR 2024: Getting older younger may be driving rise in early-onset cancers

As with most common diseases of the developed world, aging is the major risk factor for developing cancer. Most of the half-dozen hallmarks of precancer that were published last week by investigators from Vanderbilt University and the Fred Hutchinson Cancer Research Center are also hallmarks of aging. Unfortunately, scientists reported at the American Association for Cancer Research’s (AACR) 2024 annual meeting this week that accelerated aging is increasing, and may be driving an increase in early-onset cancers.
