Experimental mRNA gene therapy suppresses preeclampsia in mice

A gene therapy based on an enhanced lipid nanoparticle (LNP) loaded with vascular endothelial growth factor (VEGF) mRNA could be developed for the treatment of preeclampsia, according to a study in mice in which it alleviated maternal hypertension until the end of gestation and improved fetal health. Preeclampsia is a disease that affects 3% to 5% of pregnant women, who suffer from hypertension and proteinuria, elevated levels of protein in the urine, during pregnancy. Read More

Glutathione S-transferase inhibitor displays robust efficacy in lung cancer models

Glutathione S-transferase (GSTP) is an enzyme involved in the detoxification of several xenobiotics such as tobacco carcinogens or cisplatin drugs and is overexpressed in KRAS-mutant lung, colon and pancreatic cancer, among others. The upregulation of GSTP may partially contribute to chemotherapy and radiotherapy resistance.

Ideaya nominates KAT6/7 inhibitor as development candidate

Ideaya Biosciences Inc. has nominated IDE-251, a dual KAT6/7 inhibitor targeting 8p11 amplification tumors in breast and lung cancers, as a development candidate. IDE-251 is an equipotent, highly selective, small-molecule dual inhibitor of KAT6 and -7, both of which have been shown to support cancer cell survival. Read More

Candid Therapeutics to strengthen TCE pipeline through strategic collaborations

Candid Therapeutics Inc. has entered into a number of collaborations aimed at advancing its pipeline of T-cell engagers (TCEs) for autoimmune diseases. The company will be collaborating with Epimab Biotherapeutics Inc., Ab Studio Inc. and Nona Biosciences. Read More

ETX-197/BG-68501 shows antitumoral activity in CCNE-amplified cancers

In tumoral cells, the modulation of the G1/S phases of cell cycle is destabilized by amplification and high expression of cyclin E (CCNE) or by mutation or loss of retinoblastoma 1 (RB1) gene. Cancer cells meeting these characteristics have been sown to be highly sensitive to cyclin-dependent kinase 2 (CDK2) depletion. Read More

Verge Genomics selects new development candidate for long-term weight management

Verge Genomics (Verge Analytics Inc.) has nominated a second development candidate, VRG-201, targeting long-term weight management and metabolic homeostasis. VRG-201 is an oral, first-in-class small-molecule therapy that targets the underlying metabolic pathways leading to abnormal weight gain and the development or worsening of obesity. Read More

Crossbow reports preclinical data on first development candidate

Cathepsin G (CTSG) is overexpressed and aberrantly localized for antigen presentation on acute myeloid leukemia blasts and stem cells compared to normal hematopoietic progenitors. Earlier this year, Crossbow Therapeutics Inc. announced the nomination of its first development candidate, CBX-250, a TCR-mimetic (TCRm) bispecific T-cell engager (TCE) antibody targeting a CTSG peptide-human leukocyte antigen (pHLA) complex and CD3. Lento Bio divulges new cysteine derivatives for presbyopia

Lento Bio Inc. has synthesized cysteine derivatives reported to be useful for the treatment of presbyopia. An exemplified compound (Cpd in row 4 in tbl1 pg 20, claim 18) decreased in lens stiffness in C57BL/6 mice (at 20 mM; as eye drops). Read More
Acute myeloid leukemia

Janssen presents first-in-class bispecific Vδ2 T-cell engaging antibody

The lack of acute myeloid leukemia-specific antigens is one of the challenges for targeted immunotherapy together with on-target off-tumor toxicities due to the expression of the target in normal myeloid cells. A subset of T cells – Vδ2 T-cells – which represent ~5% of the T-cell population in healthy donors, are seen as part of emerging immunotherapeutic strategies. Read More

Iregene Therapeutics describes new BMPR1B inhibitors

Iregene Therapeutics (Suzhou) Co. Ltd. has identified pyrazolecarbonyl piperazinone compounds acting as bone morphogenetic protein receptor type-1B (BMPR1B) inhibitors reported to be useful for the treatment of cancer and pulmonary hypertension. Read More

Oncolytic viruses deliver B7H3-targeting BiTE to the tumor site, enhancing antitumor T-cell immunity

B7H3 is a pan-cancer antigen considered a promising target for immunotherapy. However, targeting B7H3 using bispecific T-cell engagers (BiTEs) presents the problem of systemic toxicity. Read More

Shanghai Institute of Organic Chemistry discovers new LRRK2 inhibitors

Shanghai Institute of Organic Chemistry has described leucine-rich repeat kinase 2 (LRRK2; dardarin) inhibitors reported to be useful for the treatment of cancer, autoimmune diseases, inflammatory disorders, Parkinson’s disease and Alzheimer’s disease, among others. Read More
