Scynexis Inc. has announced that a novel series of antifungal compounds utilizing its proprietary triterpenoid antifungal platform are among five projects funded by a federal grant awarded to the new accelerator consortium led by researchers from Hackensack Meridian Center for Discovery and Innovation (CDI) and the Johns Hopkins Bloomberg School of Public Health.
Current approved antifungal therapies are limited by their associated adverse events. SCY-247 is a new triterpenoid antifungal for intravenous or oral administration that inhibits the glucan synthase enzyme, which is a key member for the fungal cell wall.
Researchers from Scynexis Inc. have recently presented details on the preclinical characterization of SCY-247, a second-generation triterpenoid with glucan synthase inhibitor activity and intended to be used for the treatment of invasive mucormycosis.
Fearing cross contamination might have occurred during manufacturing, Scynexis Inc. is recalling Brexafemme (ibrexafungerp tablets) from the market and putting a temporary hold on the drug’s clinical trials until it knows more details.
The advance of antifungals – a class long needful of new therapies – continued with Scynexis Inc.’s potential $593 million exclusive license agreement with GSK plc for Brexafemme (ibrexafungerp), first cleared by the U.S. FDA in June 2021 and indicated for vulvovaginal candidiasis (VVC) as well as for reduction of recurrent VVC.

Hansoh will take charge of the development, regulatory approval and commercialization of the antifungal in exchange for a $10 million up-front payment and as much as $112 million in milestones.

Under the terms, Hansoh will take charge of the development, regulatory approval and commercialization of the antifungal in the region in exchange for a $10 million up- front payment and as much as $112 million in development and commercial milestones, plus low double-digit royalties on net product sales.
There are millions of fungal species, but it appears only a few hundred of them cause serious fungal infections that are responsible for an estimated 1.5 million deaths globally each year and are equally as dangerous as bacterial and viral infections. Last week, the U.S. CDC pointed the spotlight on this situation with its Fungal Disease Awareness Week.
