Study uncovers antimalarial kinase inhibitors

Chemoproteomics studies have uncovered multiple potential kinase drug targets in the Plasmodium protozoan that causes malaria, and subsequent screening of approved kinase inhibitor drugs has now thrown up several hits with lethal activity at different stages of the complex life cycle of the parasite. Researchers at the University of Cape Town, South Africa, working with scientists at Glaxosmithkline plc, describe the activity in Plasmodium of one of these kinase inhibitors, the cancer drug sapanisertib, in a paper appearing in Science Translational Medicine, Oct. 19, 2022.

GSK plc is preparing filings for its respiratory syncytial virus (RSV) vaccine in older people following supportive late-stage trial results, a product which could play a vital role as it spins out its consumer business arm. The vaccine could be a potential blockbuster for GSK, worth as much as $7 billion in peak annual sales if approved in all its potential uses – although trials in pregnant women remain on hold because of safety concerns.

GSK’S 2-dose HPV vaccine gains expanded approval in China

China has granted a green light for Glaxosmithkline plc’s human papillomavirus (HPV) vaccine, Cervarix, for girls ages 9 to 14. The HPV bivalent vaccine is used against types 16 and 18 in a two-dose course to prevent cervical cancer. It’s the first imported two-dose HPV vaccine for the age group to be approved in China, the company said. A three-dose schedule of Cervarix won Chinese approval in July 2016 for girls and women ages 9 to 25. To date, the two-dose regimen has been approved in about 100 global markets.

GSK takes Affinivax in $3.3B deal, MAPS way in pneumococcal disease

Glaxosmithkline plc (GSK) pledged $2.1 billion up front and as much as $1.2 billion in development milestone payments to take over privately held Affinivax Inc. and bring aboard the phase II-stage, next-generation, 24-valent pneumococcal vaccine candidate, AFX-3772. London-based GSK is buying all outstanding shares of Affinivax, of Cambridge, Mass., for $2.1 billion upon closing the deal in the third quarter of this year, and two potential milestone payments of $600 million to be paid if certain pediatric development goals are reached.

Sierra Oncology shares climb nearly 39% as GSK tables $1.9B offer

Shares in Sierra Oncology Inc. (NASDAQ:SRRA) surged 38.5% to $54.75 on April 13 as Glaxosmithkline plc made a $55-per-share offer for the company, which values it at $1.9 billion. The offer represents a premium of about 39% on Sierra’s April 12 closing share price of $39.52 and a premium of about 63% over its volume-weighted average share price over the last 30 trading days.

After an eight-year academic collaboration with Glaxosmithkline plc, Kynos Therapeutics Ltd. has spun out of Edinburgh University with a £9 million (US$11.7 million) financing and a phase I-ready program. The company specializes in kynurenine 3-monooxygenase (KMO), a mitochondrial outer membrane protein that is a critical gatekeeper in the kynurenine pathway of tryptophan metabolism, and which regulates inflammation and immunity.
