Mapping and categorizing cancer drug resistance mechanisms

Oct. 21, 2024
By Nuala Moran
Starting with a study of how mutations affect sensitivity to 10 molecularly targeted drugs, researchers have laid the foundations for a prospective, systematic approach to understanding the genetic mechanisms behind cancer drug resistance. These insights will inform the development of drugs that avoid resistance emerging. For existing drugs, it will be possible to better tailor treatment and to identify second-line therapies for patients whose tumors become resistant.

CROI 2024: New options, and concerns, on HIV drug resistance

March 12, 2024
By Mar de Miguel
One topic at the 31st Conference on Retroviruses and Opportunistic Infections (CROI 2024) held in Denver this month was that resistance to antiretroviral therapy (ART) has become a public health problem for people living with HIV. Without a vaccine or a cure, these patients depend on treatments that suppress viremia by preventing the virus from replicating. They are lifelong treatments and, until new advances succeed in eradicating the virus from reservoirs, the only option available.

IMPDH2 as new therapeutic target for oxaliplatin resistance in colorectal cancer

Feb. 20, 2024
Since oxaliplatin is currently one of the most effective chemotherapeutic drugs used in the treatment of colorectal cancer (CRC), resistance to oxaliplatin usually leads to therapeutic failure and poor prognosis in CRC patients. A team at The Sixth Affiliated Hospital - Sun Yat-sen University aimed to investigate the mechanisms of metabolic reprogramming in oxaliplatin resistance in CRC.

TACC3 inhibition enhances antitumor efficacy of T-DM1 in HER2-positive breast cancer

Feb. 20, 2024
In a recent study, scientists from Medical University of South Carolina aimed to evaluate whether immunogenic cell death (ICD) can trigger resistance to anticancer therapies and explore potential therapeutic opportunities to revive ICD and restore drug sensitivity.

TCF4 genetic network behind melanoma resistance to immune checkpoint blockade

Jan. 16, 2024
Although treatment outcomes have improved in metastatic melanoma since the use of immune checkpoint blockade (ICB), it still remains a medical challenge. Melanoma cells are thought to adapt several phenotypic states, such as mesenchymal-like state (MES), which may modulate their sensitivity to therapy. An international team of researchers has now investigated the mechanisms behind melanoma cells’ resistance to ICB.
